Geboren in Queens, New York, studierte Sumpter am Marymount Manhattan College, wo sie den Schwerpunkt Kommunikation legte. Sie begann ihre Karriere als Model und erschien in verschiedenen Werbespots für Produkte wie Hewlett-Packard und Liz Claiborne's Curve Parfums. Im Jahr 2004 war sie Co-Moderatorin der N-Netzwerkserie Best Friend's Date. Im folgenden Jahr bekam sie die Rolle der Layla Williamson auf One Life to Live. Seit Januar 2011 ist Sumpter auf Gossip Girl als Raina Thorpe zu sehen, die Tochter von Russell Thorpe (Michael Boatman) und Chuck Bass' Liebesinteresse.
