Sa carrière s'est étendue sur plus de 40 ans, avec des apparitions dans plus de 40 films. Elle est surtout connue pour son rôle de Mary Hatch Bailey dans le film de Frank Capra de 1946, La vie est belle. En 1953, elle a reçu l'Oscar de la meilleure actrice dans un second rôle pour son interprétation de Lorene Burke dans le drame de guerre Tant qu'il y aura des hommes. Donna Reed est également connue pour son travail à la télévision, notamment pour son rôle de Donna Stone dans la sitcom The Donna Reed Show (1958-1966). Elle a reçu de nombreuses nominations aux Emmy Awards pour ce rôle et le Golden Globe de la meilleure actrice de télévision en 1963.
