Marisa Tomei est née le 4 décembre 1964 à Brooklyn, New York, de sa mère Patricia "Addie" Tomei, professeur d'anglais et de son père Gary Tomei, avocat. Marisa a également un frère, l'acteur Adam Tomei. Enfant, la mère de Marisa corrigeait fréquemment son discours pour éliminer son lourd accent de Brooklyn. Adolescente, Marisa a fréquenté le lycée Edward R. Murrow et a obtenu son diplôme dans la classe de 1982. Elle était un an dans ses études collégiales à l'Université de Boston lorsqu'elle a abandonné pour un rôle de co-vedette dans le drame de jour CBS As the World Turns (1956). Son rôle dans cette émission a ouvert la voie à son entrée dans le cinéma: en 1984, elle a fait ses débuts avec un petit rôle dans The Flamingo Kid (1984). Trois ans plus tard, Marisa est devenue connue pour son rôle de Maggie Lawton, la colocataire à l'université de Lisa Bonet, dans la sitcom A Different World (1987).
