John Shaft is een keiharde privédetective, die te maken krijgt met de legendarische gangster Bumpy Jonas, de leider van een beruchte bende. Vervolgens krijgt hij het aan de stok met een andere groep zwarte criminelen. Maar uiteindelijk moet hij juist met deze twee groepen samenwerken als blijkt dat de dochter van Bumpy is ontvoerd door de mafia. Shaft zet alles op alles om Bumpy's dochter terug te krijgen.
