De slordige en ondeugende Deense dog Marmaduke heeft een groot hart, maar komt altijd in de problemen. Hij is het toonbeeld van de uit de kluiten gewassen schoothond, met een voorliefde voor kattenkwaad en een diep gevoel van liefde en verantwoordelijkheid voor zijn menselijke familie, de Winslows. Als een legendarische hondentrainer gelooft dat hij Marmaduke kan helpen om de eerste Deense dog in de geschiedenis te worden die de Westminster Champion-hondenshow wint, moet Marmaduke ontdekken of hij het juiste talent heeft om te schitteren.
