Его отец Дик Кьюсак (также известен как Ричард Кьюсак) занимался рекламным бизнесом, снимал документальные фильмы (и даже получил «Эмми») и сам играл в кино. Его мать Ненси — учитель математики. Брат Билл и сёстры Энн, Джоан и Сьюзи пошли по стопам отца и стали актёрами. В 8 лет поступил в театральную школу, где преподавали друзья семьи Бёрн и Джойс Пивены. В этой школе уже учились Энн, Джоан, а также сын Пивенов Джереми, который станет лучшим другом Джона на протяжении многих лет.
