Edward Gargan wurde 1902 als Sohn irischer Eltern in Brooklyn, New York, geboren. Er war der ältere Bruder des Schauspielers William Gargan, mit dem er am 17. Juli Geburtstag hatte. Edward Gargan begann seine Karriere als Musicaldarsteller am Broadway. Er sang in Musicals wie „Good News“, „Rose-Marie“ und anderen erfolgreichen Musicals der 1920er-Jahre sowie in Opern. Eine seiner frühen Rollen war „Polly of Hollywood“ im Jahr 1927. 1930 verkörperte er den Polizisten Mulligan, eine der Hauptfiguren in „Strictly Dishonorable“. 1932 ging er nach Hollywood und spielte im darauffolgenden Jahr in dem Film „David Harum“. In den folgenden 19 Jahren wirkte er in einer Vielzahl von Filmen mit. Gargan war einer der produktivsten Nebendarsteller der Filmgeschichte und spezialisierte sich auf die Darstellung von einfältigen Polizisten und begriffsstutzigen Sidekicks. Er wirkte in fast 300 Spielfilmen über einen Zeitraum von drei Jahrzehnten (1921–1952) mit und war von 1951 bis 1953 im Fernsehen zu sehen. Gargan starb 1964 in New York City.
