Dopo aver perso la moglie sette anni prima, l'eccentrico dottor John Dolittle, famoso medico e veterinario dell'Inghilterra della Regina Vittoria, vive in solitudine dietro le alte mura della sua dimora con un esercito di animali esotici a fargli compagnia. Ma quando la giovane Regina si ammala gravemente, Dolittle con riluttanza è costretto a salpare per un'epica avventura in un'isola leggendaria in cerca di una cura, ritrovando così il suo spirito e il suo coraggio mentre combatte vecchi avversari e scopre creature meravigliose.
