Sam Rockwell wurde am 5. November 1968 in San Mateo, Kalifornien, als einziges Kind von zwei Schauspielern, Pete Rockwell und Penny Hess, geboren. Die Familie zog nach New York, als er zwei Jahre alt war und lebte zuerst in der Bronx und später in Manhattan. Als Sam fünf Jahre alt war, trennten sich seine Eltern, woraufhin er und sein Vater nach San Francisco zogen, wo er später aufwuchs, während Sommer und andere Zeiten mit seiner Mutter in New York verbracht wurden. Er gab sein Schauspieldebüt, als er zehn Jahre alt war, zusammen mit seiner Mutter und besuchte später die J Eugene McAteer High School in einem Programm namens SOTA. Schon in der High School machte er seinen ersten großen Durchbruch, als er im Independent-Film Clownhouse (1989) auftrat. Die Handlung drehte sich um drei entflohene Geisteskranke, die sich als Clowns verkleiden und drei Brüder allein nach Hause terrorisieren - Sam spielte den ältesten der Brüder. Sein nächster großer Durchbruch sollte kommen, als er in einer kurzlebigen NBC-Fernsehserie namens Dream Street (1989) als Star geplant war, aber er wurde bald gefeuert. Nach dem Abitur kehrte Sam für immer nach New York zurück und absolvierte zwei Jahre lang eine private Ausbildung im William Esper Acting Studio. In dieser Zeit trat er in verschiedenen Rollen auf, wie z.B. bei den ABC Afterschool Specials (1972): Over the Limit (1990) (TV) und HBO's Lebensgeschichten: Familien in der Krise (1992): Toter Betrunkener: Die Kevin Tunell Story (Staffel 1 Episode 7: 15. März 1993); der Hauptdarsteller in Teenage Mutant Ninja Turtles (1990); und ein Gaststar in einer Emmy preisgekrönten Episode von Law & Order (1990), während er eine Reihe von regulären Jobs arbeitete und in Theaterstücken auftrat. Nach einigen kleineren Auftritten in Filmen wie Woody Allen's Celebrity (1998) und der modernen Version von A Midsummer Night's Dream (1999), in der er Francis Flute spielte, hatte er größere Rollen in zwei der größeren Hitfilme zu bieten: The Green Mile (1999) und Galaxy Quest (1999) begeistern Publikum und Kritik gleichermaßen mit seinen chamäleonartigen Auftritten als wahnsinniger Killer im ersten und alberner Schauspieler im zweiten. In jüngerer Zeit trat er in einer weiteren Reihe von Mainstream-Filmen auf, vor allem als Eric Knox in Charlie's Angels (2000) und als Zaphod Beeblebrox in The Hitchhiker's Guide to the Galaxy (2005), während er weiterhin in kleineren unabhängigen Filmen auftrat. Nach mehr als zehn Jahren im Geschäft hat Sam seinen Erfolg verdient. Im Jahr 2018 gewann er einen Oscar als bester Nebendarsteller als unruhiger Polizeibeamter in Three Billboards Outside Ebbing, Missouri (2017).
