Julianne Moore wurde am 3. Dezember 1960 in Fayetteville, North Carolina, geboren und zog in ihrer Kindheit häufig um. Die Familie lebte in Alabama, Georgia, Texas, Virginia, New Jersey, Nebraska, New York, Juneau in Alaska, dann in Panama und Deutschland. Ihr Vater war Militärrichter, ihre Mutter eine aus Schottland stammende Sozialarbeiterin. Ihre Schauspielausbildung absolvierte Julianne Moore ab dem Jahre 1983 an der Boston University School of Performing Arts, es folgten erste Bühnenauftritte in Off-Broadway-Produktionen wie Serious Money und Ice Cream – Hot Fudge, ehe Julianne Moore Ende der 1980er Jahre in TV-Produktionen wie der Serie B.L. Stryker erste Erfahrungen vor der Kamera sammelte. Ab 1992 kam Julianne Moores Kinokarriere mit einer Reihe von bemerkenswerten Nebenrollen in Gang: So etwa in Curtis Hansons Thriller Die Hand an der Wiege, anschließend in dem Thriller Auf der Flucht neben Harrison Ford und nicht zuletzt in Robert Altmans Episodenfilm Short Cuts. Es folgte für Julianne Moore eine Hauptrolle in Louis Malles Theaterverfilmung Vanya – 42. Straße sowie die erste Zusammenarbeit mit Todd Haynes in Safe als Hausfrau am Rande des Nervenzusammenbruchs. Danach stand Julianne Moore einer erfolgreichen Hollywood-Karriere nichts mehr im Wege. Julianne Moore pendelte fortan zwischen großen Mainstream-Produktionen wie Assassins – Die Killer, Vergessene Welt – Jurassic Park und Hannibal sowie künstlerisch orientierten Filmen wie Boogie Nights (der ihr unter anderem eine Oscar-Nominierung einbrachte), Magnolia und The Hours – Von Ewigkeit zu Ewigkeit. In Nine Months spielte Julianne Moore an der Seite von Hugh Grant. In Dem Himmel so fern spielte Julianne Moore 2002 zum zweiten Mal eine Hauptrolle für I’m Not There -Regisseur Todd Haynes. Trotz ihrer grandiosen Filmerfolge vergaß Julianne Moore nie ihre Liebe zum Theater. Unter anderem stand sie neben Al Pacino in Der Vater und neben Meryl Streep in An American Daughter auf der Bühne. Das Gesicht der Wahrheit, in dem sie neben Samuel L. Jackson spielt, floppte 2006. Daraufhin übernahm Julianne Moore eine Rolle in Liebe ist Nervensache unter der Regie ihres Ehemannes Bart Freundlich. In Trust the Man spielt auch der gemeinsame Sohn Caleb eine kleine Rolle. 2009 bekam sie für ihre Darstellung der alkoholsüchtigen Charlotte in A Single Man berauschende Kritiken und diverse Auszeichnungen.
